Hmmm. Gotham would be easier. Get the Bat-family out of the way and let the PC or PCs deal with the issues. Also, anyone with the DCA books has a nice selection of villains.

If you wanted to do Spider-Man villains, I'd probably set it in a town in Long Island or in the Hamptons where most of the Spdey villains hide out (Kingpin rents out a safe house there or something), and the player is the newly-minted hero in the area.
